The replay does not expire on dotabuff. It expires on Valve servers. It takes two weeks. But if you want to keep one you can download it and it will stay permanently.
In DotabuffPlus it is sorted by hero for top players of each hero (top 50) also top 80 games of the week listed. Unfortunately that's frozen at 3 months back when Valve cut them off from access to the information.

Are you referring to going to the players section of the website, viewing the top 50 list, then individually clicking each person to see what there top heroes played are? I was hoping I could sort by heroes first then by top player.
It does sort by heros first. You click Heroes. Then select a particular hero. There will be two extra tabs. One is ToP Performances, which lists the best 80 games of the week on that hero (frozen 3 months old, due to Valve adding the default private feature). The other tab is Player Rankings, which gives the a score to the top 50 players of a hero based on DBR, win rate, KDA, and matches played. Unfourtunately since DBR is included its also frozen from 3 months back.
This feature would have really been excellent had DBR been allowed to function correctly. But it does still allow you to see a list of 50 of the best pub players of each hero in the world. You can then pick out a name and go find their games to watch in the Dota 2 client.

1. How long does it take for a replay to expire? Is it after any sort of patch? I tried to watch a reply from the 15th of April only a few weeks back and the replay had already expired.
2. Is it possible to sort by Hero and then by top players? For instance, if I were interested in viewing replays of Chaos Knight I could search by that specific hero and then by the top players for that hero. I have not been able to find such functionality. Instead, I have been searching by the top players leader board, clicking individual people, and hoping I can find a hero that I am interested in that they have played recently.
